Central dogma sequencing
Given a change at one stage, predict the downstream effect on the protein.
Operon logic
Ask what happens when lactose is present or tryptophan is abundant. Inducible turns on; repressible turns off.
Mutation consequence
A frameshift is usually more damaging than a substitution. Silent mutations change nothing.
Gel electrophoresis
Smaller fragments travel further. Read the ladder before interpreting bands.
Differential expression
Same genome, different cells — the answer is always regulation, not gene loss.

Where students lose the most points

Mixing up promoter and operator
The promoter is where RNA polymerase binds; the operator is where the repressor binds. This is the classic Unit 6 trap.
Confusing inducible and repressible
lac is inducible (turned on by lactose); trp is repressible (turned off by tryptophan).
Assuming different cells have different genes
They have the same genome and express different subsets of it.
Forgetting RNA processing
Eukaryotic mRNA is spliced and capped before translation; prokaryotic mRNA is not.
Treating every mutation as harmful
Silent mutations exist, and some mutations are neutral or beneficial.
